Does a addin developed using VSTO work in outlook2000?

No the pre-requisites for VSTO 2005 are Office 2003 SP1 or greater.

Addins can work with multiple versions of Office (although MSFT doesn't recommend it). Best place to ask this would be in the Outlook newsgroups. There are quite a number of Outlook folks that create Addins - both the "traditional" kinds as well as with VSTO. They'll be the best people to give you advice on how best to approach the task.
